Retatrutide: an antidoping test for a drug no regulator approved

An antidoping lab built a test for retatrutide because amateur athletes are using a drug no regulator has approved. It stays measurable for two months.

The interesting thing about this paper is not the chemistry. It is that somebody had to write it — a laboratory built a validated assay for a drug with no legal consumer supply, because people are taking it. That is the same market researchers ordered from to see what would show up.

Why the method exists

The authors give the reason plainly: retatrutide’s effects on body composition, combined with increasing off-label use among physically active populations, raise concerns about misuse in sport. [1] Anecdotal reports among amateur athletes are cited as the prompt.

No approval exists. Whatever those athletes are injecting did not come from a prescription.

What they measured

Four users self-administered 2 mg under the skin on day one and again on day seven. Blood and urine were collected at set points afterward.

The assay detected retatrutide in plasma down to 6.14 ng/mL and would have detected it in urine down to 2.44 ng/mL. In plasma it stayed measurable for about 56 days after the first dose. In urine it never appeared at all.

That is the sample. Everything above about timing rests on four self-administered courses.

For validating an assay that is enough. For describing how long the drug stays in a population, it is not, and the 56-day figure should be read as what four people showed.

What this site will not do

Point anyone at a supply. Retatrutide is in trials, it is not sold to consumers anywhere, and this page carries no source and no dose.

The reason is not squeamishness. It is that nobody buying an unapproved peptide can establish what is in the vial — a proposition this site has already tested, both on what a copy contains when somebody assays it and on how much goes wrong between the vial and the syringe.

The part worth keeping

Fifty-six days. These molecules are engineered to persist, and a drug taken once in January is measurable in March.

That matters for anyone planning surgery, a pregnancy, or a switch between products, and it is a property of the whole class rather than of one unapproved member — including the approved ones dispensed through the two kinds of compounding pharmacy.
